カ－ドゲ－ムをしていて、カ－ドを一番多くゲットして人が勝ち～！と英語でどのように言えば自然ですか？

whoever has the most cards で「カードを一番たくさん持っている人は(誰でも）」

2) The winner is the one with the most cards at the end.
