この場合、次のような言い方ができますよ。
ーNo, not this room. It's the room at the end.

the room at the end で「奥の部屋」と言えます。
ーIt's the last room, not this one.
「ここじゃなくて、最後の（奥の）部屋だよ」
the last room で「奥の部屋」を表現できます。
手前の部屋を言うなら次のように言えます。
ーIt's the first room on the right.
「右側の手前の部屋だよ」
the first room で「最初の（手前の）部屋」
最後に on the right/left をつけると、右側/左側を言うことができます。
真ん中の部屋を言うなら次のように言えます。
ーIt's the room in the middle.
「真ん中の部屋だよ」
the room in the middle で「真ん中の部屋」
もし部屋が３つしかないなら、次の言い方もできますね。
ーIt's the second room.
「２番目の（真ん中の）部屋だよ」
