長蛇の列が出来ていたので並んでいる人に聞いてみました。

英語での聞き方はほとんど変わりがない一つの聞き方が定番となるぐらいですが、
その代表例が「Excuse me but what is this line for?」、直訳すると「あの、すいませんが、この列は何のためのものですか？」となります。
同時に、”What are you lining up for?" と聞けば「何のために並んでいるんですか？」と直接相手に聞く形でも英語ならではまた全然問題ありません。
最後は、”What is in ahead of this line?" という全く逆の発想で聞く人もいますね。訳すと、「この（列の）先には何があるんですか？」というような聞き方。
いずれの場合でも、「あの、すいません」と他人に声をかける時はどれも ”Excuse me" と始めるのが一般ですね。

line は 線、line-upは列に当たります。 「列に並びなさい」というのは line up となります。
line-up：列（名詞）
line up：列に並ぶ（動詞）
ですので、「なんのために並んでいるんですか？」と聞きたい場合は、
What are you lining up for?
となります。

If you want to ask someone about the purpose of the line, you can use any of these sentences. Although be aware the second question is more polite.
「これは何の列ですか」は、上記のように言えます。二つ目の例の方が一つ目の例よりも丁寧です。
