「間違いが３つある」と言う場合は、there are 3 differences となります。
「（間違い３つを）見つけよう」と言う場合は、Try to find them と言えます。
例文２のように一文にまとめて言うこともできます。
「これらの２つの絵に３つの間違いを見つけられるかな？」というニュアンスです。
spot は「見つける・見抜く」という意味があります。
spot の代わり find を使っても良いです。
