「最も…」は通常 the most を使って言います。
この場合、下のように言うのが自然かなと思います。
The city with the most tourists in Japan is Kyoto.
「日本で最も観光客が多い街は、京都です。」
the most tourists で「最もたくさんの観光客」
Kyoto has more tourists than anywhere else in Japan.
「京都は日本のどこよりも多く観光客がいる。」
has more tourists than anywhere else で「どこよりももっと観光客がいる」=「最も多い」を表現できます。
