この場合は次のように言う方が自然かと思います。
ーI'm two-thirds of the way through a book I'm reading.
「読んでいる本を2/3読んだ」
to be two-thirds of the way through a book で「本を2/3読んだ」と言えます。
ーI only have one-third left in the book I've been reading.
「読んでいる本があと1/3残っている」
to have one-third left で「1/3残っている」

「もう既に本の3分の2を読み終わった」
3分の１は one third
3分の２はtwo thirds
ですが、この言い方を知らなければ、パーセントに置き換えれば、言いたいことと同じ内容をもっと簡単に表現できますよ(*^_^*)
I've already read 65 percent of the book.
「本の65％を読んだ」
