「誰」と尋ねているので、whoを使うのが基本です。
in charge of ...は「…を[担当](https://eikaiwa.dmm.com/uknow/questions/36506/)している／…[担当者](https://eikaiwa.dmm.com/uknow/questions/426/)で」という意味です。

このシステムについて、誰に聞けば良いでしょうか？
Who is in charge of this system?
このシステムの担当者は誰ですか？
①Who should I go to about 〜？
〜について[尋ねる](https://eikaiwa.dmm.com/uknow/questions/51173/)にはどこに行けばいいですか？
今回社内はの話だったので、自分で足を運んで担当者のところに行くことを想定して、"go to"を使っています。

〜についてのコンタクトパーソンはどなたですか？

これだと、電話やメールでの会話としても使えます。
誰に～のことを聞いたら良いですか？と言いたい場合、
should や can を使い、聞く、尋ねるというのはaskで表現します。
誰＝who
whoの疑問詞を使って
Who should (can) I ask～? となります。
誰に聞けば良いか知りたいのですが、というニュアンスで
I would like to know who I should (can) ask～.
でも丁寧な表現で良いです。
あなたが、聞きたい用件の担当者を探している場合の表現です。
the projectの部分を言い変えればいいです。
担当をしているというのは、中心になってやっていると言うことです。
①"lead the project" または "take the lead on the project"
②"run point on the project"
②のもっと細かな表現をすると、
"This is a difficult project. Who do you have that can run point on this?"
「これは、難しいプロジェクトだ。だれか、この担当をする人がいるのかい？」
実際の担当者をお聞きする場合に、お役に立つと思います。
大きな会社では、なかなか「張本人が出て来ません」ので、細かな表現が必要です。

These are various ways in which you can ask who might be in charge at a business during a certain time. The supervisor on duty is the supervisor who is currently working during the hours in which you may need help.
上記は、担当者が誰なのか尋ねたいときに使える表現です。
The supervisor on dutyは、あなたが尋ねたい時間帯に働いている担当者のことを指します。
