’’[電話](https://eikaiwa.dmm.com/uknow/questions/51755/)に出た時’’の冒頭の決まり文句、
「[要件](https://eikaiwa.dmm.com/uknow/questions/26189/)は何ですか?」ですが、これは、
「How can I help you?」
で表せます。
「How may I help you?」
と言うこともできます。
こちらの方が少し丁寧な印象があります。
’’電話での会話中’’に、
用件がわからなくて聞きたい場合は、
下記のように相手へ伝えます。
「What is this regarding/concerning?」
「May I ask what your call is regarding/concerning?」(より丁寧)
電話での特徴的な英語表現として、
主語が「this」(この電話は)になります。
この点、注意されてください。
例を下記にお伝えします。

A:May I speak to Mr. John Smith, please?
(John Smithさんをお願いできますか?)
B:This is he./Speaking.
(はい、私です。)

''Would you mind...'' is a polite phrase... ''what is this call in connection with?''> You
want to know why this person is calling you.
May I ask why you calling?> Mayを使うことで丁寧なききかたになります。
Kindly tell me the reason for your call.> なぜ電話してきたのか理由を教えてください。と理由を尋ねる表現。
''Would you mind...'' これは丁寧な言い方です。
''what is this call in connection with?''> その人が何故電話をかけてきたのか尋ねる言い方です。
If the caller is appearing confused or just not making any sense, you could clearly ask, "What's the purpose of your call?"
"Oh, I just need to speak to Phillip."
"Sorry, he's at lunch."
Similarly, you may say: "Could you please state the reason for your call?" which is a little more formal. "Yes, I would like to make an appointment for next Monday, please."
"OK, no problem."
If the caller is appearing confused or just not making any sense, you could clearly ask, "What's the purpose of your call?"

*Hello -used as a greeting or to begin a telephone conversation.
Example -"hello there, Katie!"
* how can i help you? - trying to solve whatever issue the client has / trying to answer whatever questions the client might have.
B) How can i assist you?
*Assist -help (someone), typically by doing a share of the work.
Example -"a senior academic would assist him in his work"

In the first question, you are asking the caller how you may help him/her. It is assumed that the caller needs to be helped with either a query or would like to be transferred to another person.
There is definitely a purpose/reason for the caller to have called. So, you may politely say to the caller: Please tell me the purpose of your call.
In the third question, it is assumed that this call is about something. So, you may use the preposition 'regarding': What is your call regarding? What is your call about? What is your call concerning?
1つ目の質問では、電話の相手があなたに何をしてほしいかを聞いています。
問い合わせや他の人への取り次ぎが必要かもしれないことが想像できます。
電話の相手は目的や理由が必ずあって電話をしたのでしょう。
なので、Please tell me the purpose of your call.と丁寧に言うことができます。
3つ目の質問では、電話の用件について聞いています。regarding という前置詞を使うことができます。
「お電話は何についての用件ですか?」What is your call regarding? What is your call about? What is your call concerning?
