最も欲しいもの、の『最も』は（欲しさの度合いが最大という意味で）"the most"を使うのが自然です。
"What do you want the most right now?"と聞かれて、"Sleep."などと答えるジョークがありますね。
現実的な範囲で、何が欲しいかという質問であれば、この表現が一番よいと思います。
”What is your biggest wish?"と聞く方法もあります。"wish"は『願い』です。1つ目の表現に比べてカジュアルで、子供に対して使ったりするにはぴったりです。
『星に願いを。』的なニュアンスで、非現実的なことも連想されます。
”What would you do if..."ですが、"would"をつかって、『もしXXなら、OOする』といった仮定の表現をすることができます。"enough money and time"は、『お金と時間が十分あれば』となります。『時間とお金があったとしたら、なにしたい？』という感じです。
『今、』は"now"ですが、"at the moment"と言うこともできます。友人同士のチャットやメッセージでは"atm"と略して書くことがあります。”What are you doing atm?"『今なにしてるの？』みたいな感じです。
私はベトナム料理のフォーが好きなので、”My biggest wish at the moment is to visit Vietnam and eat pho.”
To ask what someone wants the most right now, you can ask:
What is your greatest desire right now?
Desire is what you want the most.
You can also say:
What do you long for the most?
This is quite formal as the word 'long for' means desire or to deeply want. It is a word not used as often as desire.
What do you wish for most right now?
To wish is to desire to happen or to believe can happen.
Another way to find out what someone wants is to ask them what their dream is:
What is your biggest dream?
Their dream is also what they want the most. It is used to describe a goal or wish for the future.

こちらは少し堅い言い方です。long for は、欲するとか心から欲しがっているという意味です。
long for はdesire ほどよく使われることばではありません。

Winning the Lotto and having a lot of money are this person's greatest wishes presently. Having lots of money and travelling the world is what he/she wants the most at the moment.
So, if you asked such a person the following questions:
What do you want the most at the moment?
or
What is your greatest wish presently?
You may get the following answers:
I want to have lots of money and travel the world.
or
My greatest wish at the moment is to win the Lotto and travel the world.

"I wish I won the Lotto."（宝くじで当たるといいな。）ということをよく聞いたりするでしょう。私がもし宝くじに当たったら世界中を旅したいと思います。

This could be a question relating to careers and their personal lives.

You are asking them about one thing they have always wanted but have not attained.

This is a hypothetical question that opens up a platform for a lengthy conversation.

Again this depends on the context such as who you are speaking with and if you are asking about something specific. If you are asking about their life wishes you could say "What one thing would make you happiest in your life right now?"
「誰に対しての質問なのか」、また「何か特別に欲しいものがあるのか」など状況によってどのような質問をするかが変わってきます。
夕飯について聞いているのであれば、

人生の中で何か叶えたいことがあるのかどうか聞きたいのであれば、
