You can just ask a general question
"When do you open?" "When do you close?"
"What time does your shop open?" "When do you close this evening?"
Or you can be more specific and ask if the business opens or closes at a certain time.
"Do you open at 9 am?" "Do you close at 5 pm? "
Either way is ok, it just depends on what you want to know.

By asking what a business 'opening hours' are, you have asked them to tell you the business operating hours for every day of the week.
