よく使われるスラングです。downにはおそらく①寝込んでいる②賛成する、と2つ意味があると思います。
1つ目の意味の「寝込んでいる」の用法は、
I am down with flu and high fever and have been so for a week.
「今インフルエンザで熱が出ているんだけど、ここ一週間そんな感じで寝込んでいるんだ。」
2つ目の意味の「賛成する」の用法は、
A: I'm going to Chicago this weekend. Anyone?
「今週末シカゴに行くんだけど、誰か行く？」
B: I'm down for it!
「行く！」
とこんな感じになります。このforは賛成を意味するので覚えておいてください。
To count someone in = count someone in (for something) and count someone in (on something);count someone in. to include someone as part of something.

To turn up =put in an appearance; arrive, arrive, put in an appearance,
"Half the guests failed to turn up."
To count someone in = 人や物を勘定に入れる、仲間に入れる
"Please count me in for the party."

To turn up =現れる、到着する
"Half the guests failed to turn up."
（ゲストの半分も現れなかった）
Two expressions you can use are, "I will be at the party!" or "I will be there!" Both are casual or informal expression that you would use when talking to friends or co-workers. To reply to your boss or in a more formal manner you could say, "Yes, I will be attending the party." (attending, meaning you will be at the party."
"I will be at the party!"（パーティーに行きます）
または、
"I will be there!"（行きます）
と言えます。どちらも友達や同僚に対して使えるカジュアル、インフォーマルな表現です。
相手が上司だったり、よりフォーマルに伝えたいなら、
"Yes, I will be attending the party."（はい、パーティーに行きます）
と言えます。"attending" は「〔パーティーに〕行く」という意味です。
When letting someone know you will be somewhere (at the party) you can simply say 'I'm coming!' or 'I'll be there!'

"I'll be there" is referencing that you will be going to an event that has been mentioned and queried if you would turn up or not.
