What is the population of your hometown?
Both of these answers are a good way of asking someone what their cities population is. 'Hometown' is a word we use when talking about a area that is smaller than, or not been declared a city, but this can however still be used to refer to a city in a casual manner. "Tokyo is my hometown."
どちらも、相手の住んでいる都市(city)の人口を尋ねる質問です。'Hometown' は、'City'(都市)より小さい地域について言う場合に使う言葉ですが、カジュアルな会話では 'City' を表すこともあります。

"How many people live in your city?"
This is a more casual or informal way of asking what the population of your city is. Note: you can use the word city/town/hometown also.

The word 'population' means the amount of people that live in a certain place or area so as well as using 'population' you could also ask 'how many people' these both mean the same thing
'in your city' means that city in which you live/stay in

Population' is quite a formal way of saying 'the number of people'. 'How many people live in your city?' is asking the question in more simple terms. Finally, when someone says 'your city' they are referring to the city that you live in, but this of course can be replaced by 'the city you live in'.

The three sentences you see provided above are excellent ways to ask your listener the population of the city where they live. In the second sentence you will see the word reside. This means live. This word is appropriate for formal settings, like giving a presentation at work or conducting a business meeting. It would make an awesome addition to your vocabulary.
