こういう場合の「覚悟する」はどう言えばいいのでしょうか？例えば、（人気店なので）週末は行列を覚悟しないとね、という場合、We should expect to queue on weekends. あるいは We need to be ready for a long queue on weekends. でしょうか？
We have to be prepared to stand in line on the weekend.
We should be ready to line up on weekends.
Keiさんの英語で大丈夫ですよ。queue はイギリス英語なので、アメリカ英語を使って言うと下のようになります。
例：
We have to be prepared to stand in line on the weekend.
「週末は列に並ぶのを覚悟しないとね。」
stand in line で「列に並ぶ」
We should be ready to line up on weekends.
「週末は並ぶことを覚悟しないと。」
line up でも「列に並ぶ」という意味です。
