Who are in the room? や Who is he in the room? とは言いません。
例えば、部屋に複数人いると知っていても、この場合は
ーWho is in the room?
「誰が部屋にいるの？」
と言います。
または次のようにも言えます。
ーIs there somebody in the room?
「部屋に誰かいるの？」
