様々な言い方ができると思いますが、例えば以下のような英語表現はいかがでしょうか：
When should I ask if I have any questions?
質問がある場合はいつ聞けばいいですか？
Is it OK if I interrupt you when I have a question?
質問がある場合は話を遮ってもいいですか？
interrupt は「（話を）遮る」「割り込む」のような意味です。

「いつ聞けばいいですか？」は英語で決まり文句のような表現の「When is it okay to ask a question?」を言います。
「a question」は単数形なので、代わりに複数形の「questions」を言っても構いません。
例文：
When I couldn't get what you said, when is it okay to ask a question? After you finish whatever you say? Or midway through the conversation?
僕が聞き取れない時にはいつ聞けばいいですか？一通り喋り終わってから？それとも会話の途中？
