「代わりに」は英語で「instead (of)」と言います。「人の代わりに」という文脈で「in place of」という言い方も使えます。

"in substitute"はどちらの前置詞を使われます。例えば、「砂糖の代わりに蜂蜜を使う」の場合は"use honey in substitute of sugar"または"use honey in substitute for sugar"になります。"instead"の方は前置詞を除くことができます。前置詞を除いた場合、"instead"が一番最後に来ます。
