The shop is opposite side.でも通じますが、
”The shop is the other side of the street.” （反対側） や、”The shop is right across from here.” （向かい側）の方がわかりやすい表現かもしれません。

反対側を特定する必要がありますので、the を使います。
あと、店が道路にありますので、on も必要となります。
