一番丁寧な言い方は
Is this seat taken?
このフレーズは、直接「この[席](https://eikaiwa.dmm.com/uknow/questions/70006/)は空いていますか？」と聞いていないので丁寧なのです。
直接言っても、
Is this seat available?が割と丁寧です。availableはちょっと長いのです。
Is this seat open?は一番カジュアルな言い方です。

もし空いているのなら No, go ahead. や You can use it. などと返ってくるでしょう。
空いていなければ Yeah, sorry. などと言われるかもしれません。
You could also ask the question: "Is anyone sitting here?"
1. You ask if the seat is vacant.
2. You ask if you can sit down on this seat.
"May I...." We use this construction when asking for permission to do something.
3. You ask if there is anyone using the seat. If there is someone using the seat, they may have gone away somewhere with the intention of returning shortly.
A "Is anybody sitting here?"
B "yes, my husband is. He's just gone to the bathroom."

Firstly it is always polite to say 'excuse me', especially if you are in a restaurant or if the person you would like to ask a question to is in a conversation.
'Is this seat taken?', 'Is this seat free?', 'Is there anybody sitting here?' all imply that you would like to either sit in the seat or take it away.
まずは、「excuse me」と言う丁寧です。レストランにいる場合や、会話中の相手に話しかける時は特に、です。
「Is this seat taken?」「Is this seat free?」「Is there anybody sitting here?」すべて、その席に座りたい、またはそのイスを持っていきたい、という意味合いです。
A polite way of asking someone if a seat would be "excuse me, is anyone sitting here?" by asking like this you are being considerate to the possibility that the seat is already taken. An informal quick way to ask would simply just be "is this seat taken?" This way isn't rude by any means, it's just another way of quickly asking. Both are a great way of asking.
席が空いているかどうか丁寧に尋ねるなら "Excuse me, is anyone sitting here?" と言えるでしょう。これは「人の席を取ってはいけない」という思いやりのある行動ですね。
手短に、インフォーマルに尋ねるなら：
"Is this seat taken?"
これも失礼にはなりません、ただ簡潔に聞いているだけです。
どちらを使ってもいいと思います。
