どちらも同じ意味合いになりますが”glasses on/off”は
メガネを外している・はめているというニュアンスになります。
"with/without"は日本語で言えば「あり・なし」という感じです。

The usual effect of changing from glasses to contacts is that the person looks younger. It may also be possible to compliment this person on the colour of their eyes as they are more visible now.

Sometimes we can;t see people's eyes when they are wearing glasses - contact lenses make them much more noticeable.

It is important to say 'even better', because it implies that the person looked great with the glasses but they look more beautiful without them.
If this is not said it can be considered as an insult.
Example
A: How do I look?
B: You look even better without glasses.

This way you make no reference to the way the person looked with the glasses.

If you say that something really suits them then you are saying that the change of glasses to contact lenses, really has an affect on their appearance in a positive way. You can say: "That really suits you!" about many things, not just the glasses / contact lenses.
