Jamieさんと違う言い方を書いてみましたが意味は同じです。[三連休](https://eikaiwa.dmm.com/uknow/questions/5553/)はthree day holidayですが週末を挟む場合はthree day weekendとも言います。Starting from~を入れると~から始まると言う意味になります。

3連休 = three days off, but if it is three consecutive days that fall on a typical weekend (Saturday, Sunday), you can call it a "long weekend".

To come up = occur or present itself, especially unexpectedly.

「ついに休みだ!」「やっとだぜ!」的なニュアンスでFinally!!も喜びを表すために使うことが結構あります。

主語と述語がなく、いわゆる文としての体裁は保っていませんが、このように名詞だけを言うことでそこに感情を乗せることは口語上はとてもよくあります。
日本語でも「よし、3連休!!」の一言だけで意味は十分伝わりますね。
