「交換する」の to change をいうまでも必要がありません。「新しいタオル」だけで十分です。新しいタオルをゲットするのに交換されるはずわけです。
to get は「もらう」で、could I... は丁寧にお願いするときに表現で、まとめると、Could I get new towels please?
他の交換してもらいたいものにも使えます。 Could I get new bedsheets please? Could I get a new toothbrush please? なども言えます。

The words 'new', 'clean' and 'fresh' all refer to the same type of towel - a new, clean one that hasn't been used! It is polite to say please to the member of staff, or alternatively you can say 'is it possible' at the beginning of the sentence. This also implies politeness.
To do this, you would simply go to the front desk and ask the member of staff for more towels in the way stated above. In most hotels, this will not be a problem, although they will often change the towels daily anyway.
All of these requests will get yo more towels in a hotel. The first and the third questions would be best for face to face interactions. It's common to call the front desk when you need more towels. For this, the second sentence would be best. "Send up" is a phrase that implies something will be delivered. In this case, it asks for towels to be delivered, but in a native, natural way.
