When something is 'based on' something else it means that the ideas, names or image has been used as an influence, or directly copied. For example - "The movie of Lord of The Rings is based on the book by JRR Tolkien."
Due to anime being animation based the setting can not directly be set in the town. What that means is, is that there are no actors on the street of the town acting out the story as would be in a normal movie production. So the location of the anime is influenced or based upon a town or place.
When a film/anime has been 'set' somewhere it means that a place has been used as the 'setting' (or location) for that production. For example - "The Shawshank Redemption is set in a US prison."

While Anime is a Japanese style and genre of animated film, we would sometimes refer to it as a cartoon. Though a cartoon does suggest that it is a child's animated film, which many anime are definitely not!
Depending on who you were discussing this with you would be able to say "This anime is set in my town or my hometown." Both town and hometown are correct. Town would indicate the current place that you live and hometown would be where you lived and were brought up as a child.

The two sentences you see above are great ways to express to your listener that the setting of a certain anime was your hometown. In the first sentence you will see the word set. If we are discussing where a TV show, movie, play, etc., is set it means when or where it is taking place. For example if we are watching a movie from the 1800's we can say 'This movie was set in the 1800's.'. I hope this explanation and these two sentences help you out!

One definition for the word set means to represent a particular place or time in a story or motion picture. So many times in English we would say, "This movie is set in New York in the 1920s".
So in this situation the best way to describe this would be: This anime is set in my hometown. Because it is drawn to represent your hometown and you can even mention the era of time.
