Do you mind-- から始まる質問文の場合、受諾する場合はNo, 断る場合はYesで答えます。
Do you mind if I sit here? は直訳すると「私がここに座ることを[気にします](https://eikaiwa.dmm.com/uknow/questions/44751/)か？」となるので、No, I don't mind. （いいえ、気にしません）、または Yes, I mind. （はい。気にします）が返答になります。
Do you mind if I sit here? と聞いて返事がNoでも断られた訳では無いのでご安心下さい！
May I sit here? は「はい」なら普通にYes,「いいえ」ならNoで答えます。

★ 語句と表現
・move over
「移動する」の move と「別の場所へ、離れた場所へ」という意味の副詞の over を合わせて「席を詰める」という意味になります。でもめっちゃ離れた場所に移動しろという意味ではないのでご安心を！ｗ
・a little
「少し」という意味で、上記と合わせて move over a little とすると「少し席を詰める」という意味になります。

一方で、席が空いていて、隣の乗客がカバンや持ち物を置いていることがあります。そういう場合は、席を指差して、Excuse me（すみません）と言って、そこ（人の持ち物が置いてある場所）に座りたいことを伝えます。
Is this seat taken? で使われている take は、「場所を占める、取る」という意味の take です。
例えば、Sit down, please. よりも丁寧な言い方として使う、Please take/have a seat. （どうぞお座りください）も、この意味の take 用いています。
英訳例は、「この席は（既に誰かに）取られていますか？」という意味になるので、「この席は空いていますか？」という意味で使えます。
